Application of xylanases from Amazon Forest fungal species in bleaching of eucalyptus kraft pulps BABT

Crude xylanase preparations from Penicillium corylophilum, Aspergillus niger and Trichoderma longibrachiatum were used to treat Eucalyptus kraft pulp, prior to chlorine dioxide and alkaline bleaching sequences. The enzyme pretreatment improved brightness and delignification of non-delignified and oxygen-bleached samples of eucalyptus kraft pulp. Xylanase preparations from T. longibrachiatum and P. corylophilum were more effective to reduce pulp kappa number. A small reduction in viscosity was obtained when the oxygen-bleached pulp was treated with xylanase preparation from A. niger. Activities of cell wall hydrolases of Euphorbia heterophylla L. (wild poinsettia) endosperm were investigated during pre- and post-emergence periods, defined as the time interval before and after 2.2 days from the start of imbibition, respectively. The activities of endo-beta-mannanase and beta-mannosidase are higher over the pre-emergence when compared to the post-emergence period and they may be involved in the process of germination in E. heterophylla. Cellulase and xylanase productions by isolated Amazon Bacillus strains using soybean industrial residue based solid-state cultivation BJM

In Brazil, a large amount of a fibrous residue is generated as result of soybean (Glycine max) protein production. This material, which is rich in hemicellulose and cellulose, can be used in solid state cultivations for the production of valuable metabolites and enzymes. In this work, we studied the bioconversion of this residue by bacteria strains isolated from water and soil collected in the Amazon region. Five strains among 87 isolated bacteria selected for their ability to produce either celullases or xylanases were cultivated on the aforementioned residue. Cloning of a novel xylanase gene from a newly isolated Fusarium sp. Q7-31 and its expression in Escherichia coli BJM

A strain of Q7-31 was isolated from Qinghai-Tibet Plateau and was identified as Fusarium sp. based on its morphological characteristics and ITS rDNA gene sequence analysis. It has the highest capacity of degrading cell wall activity compared with other 11 strains. To do research on its xylanase activity of Fusarium sp. Q7-31 while the degrading the rice cell walls, the complete gene xyn8 that encodes endo-1, 4-β-xylanase secreted by Fusarium sp. Q7-31 was cloned and sequenced. The coding region of the gene is separated by two introns of 56bp and 55bp. It encodes 230 amino acid residues of a protein with a calculated molecular weight of 25.7 kDa. Coexpression of rumen fungal xylanase and bifunctional cellulase genes in Escherichia coli BABT

ABSTRACT Rumen fungi inhabit the gastro-intestinal tract of ruminants and the most non-ruminant herbivores. Rumen fungi produce highly active plant cell wall degrading enzymes, therefore they have gained scientific interest. In this study, genes encoding xylanase (xynA-7) and cellulase (celA-5) were amplified from Neocallimastix sp. GMLF7 and Orpinomyces sp. GMLF5, respectively, and expressed in Escherichia coli. XynA-7 was found to be active only on xylan, however CelA-5 had activity both on carboxymethyl cellulose and lichenan. Comparative study for the kinetics of extracellular xylanases from Trichoderma harzianum and Chaetomium thermophilum Electron. J. Biotechnol.

Xylanases assume special importance in the paper and pulp industry as they replace toxic chemicals such as chlorine and chlorine dioxide for developing eco-friendly processes. This study evaluated xylanases produced by two fungi, the mesophilic fungus Trichoderma harzianum and a thermophilic fungus Chaetomium thermophilum. Among the polymeric substrates studied for xylanase production by both the fungi, birch wood xylan was found to be the best inducer of xylanases. Xylanases induction was subject to glucose repression. Two metabolism assays were carried out to determine corn and soybean meal metabolizable energy when enzymes were added. In the first trial, 35 cockerels per studied feedstuff (corn and soybean meal) were distributed in a completely randomized experimental design with four treatments of seven replicates of one bird each. ABSTRACT The present study was conducted to evaluate the performance, carcass yield, and histomorphometry of the small intestine of broilers fed yeast sugarcane supplemented with enzymatic blend from 22 to 42 days of age. Seven hundred broiler chicks were distributed in a completely randomized design in a factorial design (2x3+1), two levels of the enzyme blend (0 and 200 g/ton), three protein levels (0%, 6% and 12%) and a control diet. The level of 6% yeast showed higher feed intake and weight gain after 33 days. This work is aimed to produce endoglucanase through solid state fermentation in a packed bed bioreactor with the use of the fungus Myceliophtora sp. I-1D3busing a mixture of wheat bran (WB) and sugar cane bagasse (SCB) as culture medium. Preliminary tests were performed in polypropylene plastic bags, controlling the variables temperature (40, 45, and 50ºC), initial moisture content (75, 80, and 85%, w.b.), and weight proportion SCB/WB (1:1, 7:3, and 9:1). Enzymatic programs for broilers BABT

Two trials were conducted on floor pens to evaluate the performance of broilers fed with diets contained different carbohydrases enzyme programs at different ages. Trial 1- The body weight gain (BWG) was affected by the Negative Control (NC) treatment (P<0.05). However, when the birds were fed with the NC diet + Amilase (AMY)+Xilanase (XYL), the BWG improved and reached the same level as the Positive Control (PC). Trial 2- The birds received enzymes supplementation in the total phase and others only in the grower phase.The ME reduction by 120 kcal/kg increased the feed conversion ratio (FCR) (P<0.05) when compared to PC and none of the enzyme programs was able to recover the performance. El presente trabajo tuvo como objetivo reducir y/u optimizar la concentración de algunos nutrimentos en las dietas de cerdos de engorda que puede resultar benéfica en la formulación de raciones. Se realizaron 3 experimentos para evaluar el nivel de lisina, el uso de alcaloides y xilanasas en dietas para cerdos de engorda. En el experimento 1 se determinaron los niveles óptimos biológicos (NOB) de lisina total para cerdos en las etapas de crecimiento y finalización en el comportamiento productivo, las características de la canal, y la concentración de urea en plasma. ABSTRACT The objective of this study was to evaluate the effects of supplementation of two different enzymatic complexes, fibrolytic (NSPases) or amylolytic (EXP3066) enzymes, on growth performance, digestibility, behavior, and carcass characteristics of feedlot cattle. Thirty-six ½ Angus yearling bulls with an average initial weight of 391±5.0 kg were used in this experiment blocked by weight in a completely randomized design divided in three treatments: control, NSPases, and EXP3066. The basal diet was composed of 85% whole corn grain and 15% nucleus. NSPases increased average daily gain on days 63 and 84, and gain:feed ratio on days 42, 63, and 84. Xylanase activity was isolated from crude extracts of Trichoderma harzianum strains C and 4 grown at 28oC in a solid medium containing wheat bran as the carbon source. Enzyme activity was demonstrable in the permeate after ultrafiltration of the crude extracts using an Amicon system. Hyperexpression of two Aspergillus niger xylanase genes in Escherichia coli and characterization of the gene products BJM

The analysis of individual gene product should enable to clarify the role of a particular enzyme in a complex xylanase system of A. niger. The two genes encoding precursors of co-produced endo-1,4-β-D-xylanases, xynA1 and xynB, were isolated from Aspergillus niger SCTCC 400264 (SCTCC, China) by using RT-PCR technique and then successfully expressed in Escherichia coli BL21. Improvement of xylanase production by Cochliobolus sativus in solid state fermentation BJM

The xylanase production by Cochliobolus sativus strain Cs6 was improved under solid state fermentation (SSF). High xylanase activity (1079 U/g) was obtained when wheat straw was used after 8 days of incubation. Combinations of sodium nitrate with peptone or yeast extract resulted in an increased xylanase production (1543 and 1483 U/g, respectively). The Cs6 strain grown in SSF in a simple medium, proved to be a promising microorganism for xylanase production.

Induction of xylanase in thermophilic fungi Scytalidium thermophilum and Sporotrichum thermophile BABT

Regulation of xylanase production in two thermophilic fungi Scytalidium thermophilum and Sporotrichum thermophile was investigated. The expression of xylanase was found to be inducible in both the cases. Various carbon sources were tested so as to identify the inducers. Soy flour and oat spelt xylan induced maximum level of xylanase in Scytalidium thermophilum and Sporotrichum thermophile respectively. Induction of xylanase in Scytalidium thermophilum led to simultaneous induction of cellulase. Influence of agitation speeds and aeration rates on the Xylanase activity of Aspergillus niger SS7 BABT

In this study, the effect of agitation and aeration rates on xylanase activity of Aspergillus niger SS7 in 3-litre stirred tank bioreactor was investigated. The agitation rates tested were 100, 200 and 300 rpm at each airflow rates of 0.5, 1.0 and 1.5 vvm. The maximum xylanase activity in mono- agitator system was at the agitation speed of 200 rpm and aeration rate of 1.0 vvm. In bi-agitator system, at low agitation speed (100 rpm), the xylanase activity was enhanced by 13% compared to mono- agitator system for an aeration rate of 1.0 vvm. Xylanase productivity in continuous culture was higher by approximately 3.5 times than in batch culture.

Investigation of factors affecting xylanase activity from Trichoderma harzianum 1073 D3 BABT

In this study, some physiological conditions affecting the activity of xylanase enzyme produced from Trichoderma harzianum 1073 D3 were determined. In addition, stabilization of pH and temperature in liquid and semi-solid state cultivation media were investigated. It was concluded that for maximum xylanase activity, incubation at 60°C in an enzyme incubation medium with pH 5 that contained 1 % xylan was appropriate. The stability studies showed that the enzyme was relatively stable in the pH range 3-7 and retained more than 50 % of its original activity after four months.

Molecular characterization of a Xylanase-producing fungus isolated from fouled soil BJM
